Turmeric-based drug effective on Alzheimer flies

ScienceDaily (Feb. 14, 2012) ? Curcumin, a substance extracted from turmeric, prolongs life and enhances activity of fruit flies with a nervous disorder similar to Alzheimers. The study conducted at Link?ping University, indicates that it is the initial stages of fibril formation and fragments of the amyloid fibrils that are most toxic to neurons.

Ina Caesar, as the lead author, has published the results of the study in the journal PLoS One.

For several years curcumin has been studied as a possible drug candidate to combat Alzheimer’s disease, which is characterized by the accumulation of sticky amyloid-beta and Tau protein fibres. Link?ping researchers wanted to investigate how the substance affected transgenic fruit flies (Drosophila melanogaster), which developed evident Alzheimer’s symptoms. The fruit fly is increasingly used as a model for neurodegenerative diseases.

Five groups of diseased flies with different genetic manipulations were administered curcumin. They lived up to 75 % longer and maintained their mobility longer than the sick flies that did not receive the substance.

However, the scientists saw no decrease of amyloid in the brain or eyes. Curcumin did not dissolve the amyloid plaque; on the contrary it accelerated the formation of fibres by reducing the amount of their precursor forms, known as oligomers.

“The results confirm our belief that it is the oligomers that are most harmful to the nerve cells,” says Professor Per Hammarstrom, who led the study.

“We now see that small molecules in an animal model can influence the amyloid form. To our knowledge the encapsulation of oligomers is a new and exciting treatment strategy,” he said.

Several theories have been established about how oligomers can instigate the disease process. According to one hypothesis, they become trapped at synapses, inhibiting nerve impulse signals. Others claim that they cause cell death by puncturing the cell membrane.

Curcumin is extracted from the root of herbaceous plant turmeric and has been used as medicine for thousands of years. More recently, it has been tested against pain, thrombosis and cancer.

No Ferguson extradition over Turkey film: UK source (Reuters)

LONDON (Reuters) ? There is no prospect of Sarah Ferguson, the Duchess of York, being extradited to Turkey to face criminal charges over her undercover reporting for a TV documentary on Turkish orphanages, a British interior ministry source said on Friday.

Turkey sought Ferguson’s extradition after a Turkish court accused her of “breaking the law in acquiring footage and violating the privacy of five children” while making the documentary in 2008, Turkey’s Anatolian news agency said.

The charges carry a maximum jail term of 22 years six months.

Ferguson, the former wife of Queen Elizabeth’s second son Prince Andrew, posed as a potential charitable donor to visit state orphanages in Turkey and Romania, followed by hidden cameras, to highlight poor conditions at rehabilitation centers for mentally handicapped children.

The documentary, made by British channel ITV’s “Tonight” program, reported cases of children being tied to beds and disabled children being neglected by overworked staff.

Ferguson later apologized for any embarrassment the documentary might have caused Turkey.

Britain’s Home Office (interior ministry) said in a statement “The Home Office can confirm it has received a formal request for mutual legal assistance concerning Sarah, Duchess of York. It is not appropriate to comment further.”

A ministry source said there was no question of the Duchess being extradited to Turkey. “It has to be an offence in both the countries’ laws. It’s not an offence in UK law so the duchess won’t be extradited,” the source told Reuters.

Asked about the possibility of extradition to Turkey, Ferguson told Reuters in 2009: “I am a human rights activist for children. I’m apolitical and multi-faith. I go with the governments, not against them.

“But I will be a foghorn for silent whisperers if I feel that children are not given the life that they should be given. In other words if they’re in institutions and haven’t even seen the light of day, that doesn’t sit well with me.”

The poor conditions in state orphanages and psychiatric centers in Turkey have been the subject of several domestic and international investigations.

At the time, Turkish officials accused Ferguson of smearing Turkey’s image with the aim of increasing opposition to Ankara’s bid to join the European Union.

Predominantly Muslim Turkey began EU membership negotiations in October 2005, but talks have slowed to a halt because of opposition in some member states and what the EU says is lack of reforms on human rights, freedom of expression and other areas.

NEW YORK ? Jorge Posada is planning to retire after 17 seasons with the New York Yankees rather than pursue opportunities with other teams.

A person familiar with the decision told The Associated Press on Saturday that the five-time All-Star catcher will announce his retirement this month.

The person spoke on condition of anonymity because the decision has not yet been made public. Posada’s plans were first reported by sports radio station WFAN.

The 40-year-old Posada won five World Series titles with the team that drafted him in the 24th round of the 1990 draft. He became a free agent after a trying season in New York, the final year of a four-year, $52 million contract.

Earlier this offseason, Posada acknowledged that his career with the Yankees was over, but said he had offers from several other teams.

Posada’s retirement leaves shortstop Derek Jeter and closer Mariano Rivera as the two remaining players from the core group that led the Yankees to four World Series championships from 1996-2000 and one more in `09. Andy Pettitte retired after the 2010 season.

Only Bill Dickey and Yogi Berra have caught more games in pinstripes than Posada (1,574).

But Posada lost his catching job before the start of last season and struggled early in his new role as the designated hitter. The switch-hitter was batting .165 against left-handers on May 14 when he was dropped to No. 9 in the batting order and asked out of the game against Boston.

He finished batting a career-low .235 with 14 homers and 44 RBIs in 115 games, playing sporadically in September after top prospect Jesus Montero was called up. Posada did have the winning hit ? a pinch-hit, two-run single ? against Tampa Bay on Sept. 21, clinching the AL East for New York.

Posada then hit .429 in the five-game division series loss against Detroit, receiving several long ovations and chants of “Jor-ge! Jor-ge!” at Yankee Stadium.

For his career Posada hit .273 with 275 home runs and 1,065 RBIs, winning the Silver Slugger Award five times as best hitting catcher in the American League.
